The dissertation presents a comprehensive study of feline infectious peritonitis (FIP), a disease associated with infection by feline coronavirus (FCoV). The relevance of this research is determined by the widespread circulation of FCoV in cat populations, the need to improve FIP diagnosis, and the importance of providing scientific evidence for antiviral treatment with the nucleoside analogue GS-441524 and molnupiravir. The aim of the study was to investigate the epidemiological characteristics of feline coronavirus infection in selected regions of Ukraine, improve the comprehensive diagnosis of FIP in living animals, and provide scientific evidence for antiviral treatment based on clinical, laboratory, morphological, and molecular genetic investigations. The study used epidemiological, clinical, hematological, biochemical, serological, immunochromatographic, instrumental, cytological, pathological, histological, molecular genetic, and statistical methods. A total of 234 serum samples were tested by enzyme-linked immunosorbent assay (ELISA), 31 serum and fecal samples by immunochromatographic assay (ICA), and 22 effusion samples by polymerase chain reaction (PCR). Questionnaires were completed by 29 cat owners and 41 veterinarians. Morphological examination of seven effusion samples, postmortem examination of three cats, and sequencing of two PCR amplicons were performed. In addition, 9,671 laboratory test records and 368 clinical case records were retrospectively analyzed. A retrospective analysis of 6,377 laboratory tests performed at BALD Laboratory (Kyiv, Ukraine) showed that IgG antibodies against FCoV were detected in 2,178 of 5,442 cases (40.0%), while FCoV RNA was detected in 245 of 729 fecal samples (33.6%). In a cross-sectional study of 234 clinically healthy stray and shelter cats from Chernihiv, Sumy, Uman, Poltava, and Mykolaiv, the overall seropositivity rate was 58.9% (138/234; 95% CI: 52.6–65.1%). Seropositivity varied significantly between cities, ranging from 20.0% in Sumy to 80.0% in Chernihiv (χ² = 40.41; p < 0.001). A statistically significant association was found between seropositivity and keeping multiple cats in the same household (p = 0.0152). The high frequency of antibodies in clinically healthy cats indicates widespread FCoV circulation in Ukraine and confirms that FCoV infection alone is insufficient for the development of FIP. A retrospective analysis of 85 FIP cases at Zoolux Veterinary Clinic (Kyiv, Ukraine) identified 22 cats with the effusive form and 63 with the non-effusive form, including neurological and ocular manifestations. Most hematological and biochemical changes were nonspecific. However, the albumin-to-globulin (A/G) ratio and serum amyloid A (SAA) concentration were more informative. The mean A/G ratio was 0.46 ± 0.22, while the mean SAA concentration in a subgroup of 26 cats was 95.9 ± 81.9 µg/mL. In a separate clinical series of 60 cats at the OMU University Veterinary Hospital (Samsun, Türkiye), the A/G ratio at diagnosis was 0.42 ± 0.11. These findings confirm the need for a comprehensive assessment of clinical and laboratory parameters when diagnosing FIP. A retrospective analysis of 135 cases involving fluid accumulation in body cavities showed that FIP was definitively diagnosed in only 13 cats (9.6%), whereas 90.4% of cases had other causes. Among 22 cats with effusive FIP, the Rivalta test was positive in 77.3% of cases, and the A/G ratio in effusion fluid was 0.45 ± 0.23. Cytological examination of 15 effusion samples revealed findings compatible with FIP. In an additional study of four cats initially suspected of having FIP, cytological signs of sterile inflammation were identified in one case, while bacteria or evidence of neoplasia were found in the remaining samples. These results support the use of effusion cytology primarily for the differential diagnosis of FIP. Molecular genetic testing using nested PCR targeting the conserved 3′ untranslated region (3′-UTR) of the FCoV genome detected viral RNA in 12 of 22 effusion samples (54.5%). Eleven positive samples were obtained from cats with suspected effusive FIP, and one was obtained from a cat with a different initial diagnosis. Subgenomic mRNA of the FCoV M gene was detected in all 12 positive samples as a marker of viral transcriptional activity. This finding strengthens the suspicion of FIP but does not demonstrate viral replication exclusively in macrophages. An S gene fragment characteristic of FCoV type I was amplified in all positive samples. Sanger sequencing produced two nucleotide sequences deposited in GenBank under accession numbers PZ810242 (FCoV Kyiv-1) and PZ810243 (FCoV Kyiv-2). Their nucleotide identity was 90% (289/320), and the Kimura two-parameter (K2P) genetic distance was 0.1364 ± 0.0386 substitutions per site, indicating genetic diversity among local FCoV type I variants. The effectiveness of an 84-day treatment course with GS-441524-based preparations was evaluated retrospectively in 88 cats treated at Zoolux Veterinary Clinic. Complete recovery was recorded in 69 cats (78.4%), death in 12 (13.6%), and relapse in seven (8.0%). By day 14, clinical improvement was observed in 87.5% of cats. By day 84, the A/G ratio had increased significantly from 0.32 ± 0.11 to 0.81 ± 0.16 (p < 0.001), while SAA concentration had decreased from 108.0 ± 39.0 to 14.6 ± 7.3 mg/L (p < 0.001). An initial A/G ratio above 0.40 was associated with a higher probability of recovery (OR = 3.7; p < 0.01). A positive clinical response during the first seven days was also associated with a favorable treatment outcome (p < 0.001). Changes in the A/G ratio and SAA concentration are therefore useful for monitoring treatment effectiveness. The effectiveness of molnupiravir was investigated retrospectively using data from 60 clinical FIP cases at the OMU University Veterinary Hospital (Türkiye). During a 60-day course of molnupiravir treatment, survival remained at 94.3% at all assessment points. In contrast, survival among cats receiving conventional treatment with prednisolone and pentoxifylline decreased from 60.0% on day
